This ELISA kit is designed for the quantitative detection of Human Ubiquitin C-terminal Hydrolase L1 (UCH-L1) in human serum, plasma, and other related biological samples. The method employed is a double-antibody sandwich ELISA, which ensures high specificity and sensitivity for UCH-L1 measurement.
The principle of the assay involves coating microtiter plate wells with a specific monoclonal antibody against UCH-L1. After sample incubation, the target protein binds to the immobilized antibody. A horseradish peroxidase (HRP)-labeled secondary antibody is then added, forming a complex of antibody-antigen-enzyme-labeled antibody. Following thorough washing steps, the TMB substrate is introduced, and the enzyme reaction leads to a color change from blue to yellow upon acid termination. The intensity of the color is directly proportional to the concentration of UCH-L1 in the sample.
The absorbance is measured at 450 nm using a microplate reader, and the UCH-L1 concentration is determined by comparing the sample OD values to a pre-established standard curve. This technique provides accurate and reproducible results, making it suitable for clinical and research applications.

Sample Requirements:
It is recommended to process the samples immediately after collection. If storage is necessary, keep the specimens at -20°C and avoid repeated freeze-thaw cycles. Samples containing sodium azide (NaN3) should not be used, as this compound can inhibit HRP activity and interfere with the assay. Always follow proper handling and storage procedures to ensure reliable results.
